摘要:
目的:观察孙氏头腹针对焦虑性失眠大鼠BDNF-TrkB信号通路表达的影响。方法:将SD大鼠随机分为空白组、模型组、常规针刺组、头腹针刺组。除空白组外,其他三组采用睡眠剥夺、焦虑型心理应激法结合腹腔注射对氯苯丙氨酸法联合复制焦虑性失眠大鼠模型。常规针刺组进行常规穴位针刺治疗,头腹针刺组给予头针与腹针联合治疗,头腹针刺组在头部穴位进行电针干预,频率为2 Hz连续波,电流约0.5 mA,每日1次,持续时间30 min,连续干预1周。比较各组大鼠的戊巴比妥钠协同睡眠实验、旷场试验(OFT)等行为学指标。采用蛋白质印迹法(Western blotting,WB)与实时荧光定量聚合酶链式反应(Quantitative real-time polymerase chain reaction,qPCR)检测海马BDNF-TrkB的mRNA与蛋白表达水平。结果:与空白组相比,模型组的睡眠潜伏期(SL)延长、睡眠持续时间(ST)缩短,水平运动距离与垂直探索次数均减少,说明造模成功。与模型组相比,两针刺组的SL缩短、ST延长,水平运动距离与垂直探索次数均增多(均P<0.05)。与常规针刺组相比,头腹针刺组的SL缩短、垂直探索次数增多(均P<0.05)。与空白组相比,模型组BDNF、TrkB的mRNA与蛋白表达降低(均P<0.01)。与模型组相比,两针刺组BDNF、TrkB的mRNA与蛋白表达增多(均P<0.05)。与常规针刺组相比,头腹针刺组BDNF的mRNA表达增多、BDNF、TrkB的蛋白表达增多(P<0.05)。结论:孙氏头腹针与常规针刺均能改善焦虑性失眠大鼠的睡眠质量和焦虑样状态,促进BDNF、TrkB的mRNA与蛋白表达,其中孙氏头腹针的疗效更优,为临床治疗焦虑性失眠提供新的治疗思路。
Abstract:
Objective:To observe effect of Sun’s head-abdominal acupuncture on the BDNF-TrkB signaling pathway expression in rats with anxiety-induced insomnia.Methods:SD rats were randomly divided into control group,model group,conventional acupuncture group,and head-abdominal acupuncture group.Except for the control group,anxiety-insomnia models were established in the other three groups using sleep deprivation,anxiety-inducing psychological stress,and intraperitoneal injection of para-chlorophenylalanine.The conventional acupuncture group received standard acupoint treatment,while the head-abdominal acupuncture group received combined head and abdominal acupuncture with electroacupuncture (2 Hz continuous wave,0.5 mA) applied to head acupoints,30 minutes daily over one week.Behavioral assessments,including the pentobarbital-induced sleep test and open field test (OFT),were conducted.Additionally,Western blot (WB) and quantitative polymerase chain reaction (qPCR) were employed to detect hippocampal BDNF-TrkB mRNA and protein expression levels.Results:Compared to the control group,the model group exhibited prolonged sleep latency (SL),shortened sleep duration (ST) (P<0.01),and reduced horizontal/vertical movement in OFT (all P<0.05),confirming successful modeling.Both acupuncture groups showed shortened SL,prolonged ST (P<0.01),and increased motor activity (P<0.05) compared to the model group.The head-abdominal acupuncture group further outperformed the conventional group in SL reduction and vertical exploration (P<0.05).Compared with the control group,the model group showed significantly decreased mRNA and protein expression of BDNF and TrkB (P<0.01).Both acupuncture groups exhibited upregulated BDNF-TrkB expression (P<0.05),with the head-abdominal group showing higher BDNF and TrkB protein levels than the conventional group (P<0.05).Conclusion:Both Sun’s head-abdominal acupuncture and conventional acupuncture can improve sleep quality,alleviate anxiety-like behaviors,and enhance BDNF-TrkB signaling in anxiety-insomnia rats.Notably,Sun’s head-abdominal acupuncture shows superior therapeutic efficacy,providing a novel clinical treatment strategy for anxiety-induced insomnia.
