摘要:
摘 要 目的:通过观察电针百会穴对创伤后应激障碍(PTSD)大鼠睡眠时相和下丘脑细胞因子TNFα、IL1β、IL4和IL10含量水平的影响研究,探讨其治疗PTSD所致失眠可能的作用机制。方法:选取30只清洁级健康SD雄性大鼠随机分为空白组、模型组和电针组,每组10只。通过手术脑内植入微型电极,恢复15 d后,利用单程长时应激法(SPS)复制PTSD动物模型,模型复制后单笼饲养7 d,电针组同时给予针刺百会穴治疗。监测各组大鼠8 h脑电和肌电后断头取脑,取下丘脑组织,低温匀浆,Elisa方法测定其中IL1β、TNFα、IL4和IL10含量的水平。结果:与空白组比较,模型组大鼠快速眼球运动睡眠(REMS)、漫波睡眠浅睡期(SWS1)和慢波睡眠深睡期 (SWS2)时间均明显缩短(P<0.05),IL1β和TNFα水平显著降低(P<0.05)、IL4含量明显升高(P<0.05)、IL10变化不明显(P>0.05)。与模型组比较,电针组大鼠SWS1、REMS时长、TNFα和IL4水平明显增加(P<0.05),IL4含量显著降低 (P<0.05),SWS2、IL1β和IL10虽有改变的趋势,但差异无统计学意义(P>0.05)。 结论:电针百会穴可以改善PTSD失眠大鼠睡眠时相,其作用机制与调节下丘脑TNFα、IL4的水平有关。
Abstract:
Abstract 〖HT5”SS〗Objective:To investigate the effects of electrical acupuncturing in Baihui acupoint on sleep phases of posttraumatic stress disorder (PTSD) rats and the level of IL1β、TNFα、IL4 and IL10 in the hypothalamus. Methods:30 clean grade healthy SD male rats were randomly divided into the vehicle group,the model group and the electroacupuncture group,10 in each group. Microelectrode were implanted in brain of each rat. After 15 days of recovery,a single prolonged stress (SPS) method was used to replicate the PTSD animal model. After the model was replicated,the rats were fed in a single cage for 7 days. The electroacupuncture group was treated with acupuncture at Baihui point at the same time. Monitoring of 8hour EEG and EMG in each group of rats,Severed head,took off hypothalamus tissue and homogenize at low temperature,the levels of IL1β、TNFα、IL4 and IL10 were evaluated by Elisa. Results:Compared with vehicle group,duration of SWS1,SWS2 and REMS in model group was obviously shortened(P<0.05). The level of IL4 increased(P<0.05) markedly,while there was no significant change of IL10(P>0.05). Compared with model group,the duration of SWS1 and REMS and the level of TNFα and IL4 increased dramatically and IL4 decreased obviously in electroacupuncture group(P<0.05). There was no significant change of SWS2、IL1β and IL10 in electroacupuncture group(P>0.05). Conclusion:Electrical acupuncturing Baihui acupoint can improve the sleep quality of PTSD rats. The mechanism is related to regulate the level of TNFα and IL4 in the hypothalamus. 
