摘要:
目的:探讨针刺百会、大椎对脑缺血模型大鼠GAP-43表达的影响,探索其对神经功能再生的影响机制,为临床针刺百会、大椎治疗缺血性脑血管病提供实验依据。方法:选取SPF级雄性大鼠150只,体质量230~270 g,采用随机数字法随机分为五个大组,每组30只大鼠。各组又随机平均分为术后3 d、7 d、21 d 3个不同时段亚组,每组10只。采用线栓法制备脑缺血模型。电针组针刺主穴后,接G6805-2型电针仪,每天1次,每次30 min。针刺组予针刺干预,不接电针,每5 min行针1次,每天1次,电针结合传统针刺组,行电针及传统针刺方法,每天1次。模型组术后不予处理,假手术组使实验大鼠大脑中动脉暴露,不予栓线。各组大鼠在造模后3 d、7 d、21 d 3个不同时段采用神经功能缺损评分及免疫组化法检测脑缺血周围组织GAP-43蛋白表达情况,采用JD801形态学图像分析系统进行分析,对结果进行统计学分析。结果:各时间段模型组神经功能评分与假手术组比较显著升高,差异具有统计学意义(P<0.05); 在各时间段其他组与模型组比较,电针组、传统针刺组、电针结合传统针刺组神经功能缺损评分均较模型组降低,差异具有统计学意义(P<0.05); 术后第7天和第21天,与电针组、传统针刺组比较,电针结合传统针刺组大鼠神经功能缺损评分降低,差异具有统计学意义(P<0.05)。各时间段模型组脑组织GAP-43蛋白的免疫表达与假手术组比较显著升高,差异具有统计学意义(P<0.05)。在各时间段其他组与模型组比较,电针组、传统针刺组、电针结合传统针刺组大鼠相应区域GAP-43免疫活性较模型组增高,差异具有统计学意义(P<0.05); 术后第3天、第7天和第21天,与电针组、传统针刺组比较,电针结合传统针刺大鼠GAP-43免疫活性显著增高,差异具有统计学意义(P<0.05)。结论:电针结合传统针刺百会、大椎能促进各时间点脑缺血大鼠脑组织内GAP-43表达,改善脑血管的缺血状态。
Abstract:
Objective:To investigate the effect of acupuncture Baihui(DU 20)and Dazhui(DU 14)on the expression of GAP-43 in rats with cerebral ischemia,and to explore the mechanism of its effect on nerve function regeneration,so as to provide experimental basis for clinical acupuncture of Baihui(DU 20)and Dazhui(DU 14)in the treatment of ischemic cerebrovascular disease.Methods:150 male rats of SPF grade,weighing 230-270 g,were randomly divided into 5 groups with 30 rats in each group.Each group was randomly divided into three subgroups:3 days,7 days and 21 days after operation,with 10 rats in each group.The model of cerebral ischemia was established by thread thrombus method.After acupuncture at the main point in the acupuncture group,the G6805-2 electroacupuncture instrument was connected with each once a day,30 minutes each time.Acupuncture group was treated with acupuncture intervention,acupuncture was given once every 5 minutes,once a day,acupuncture combined with traditional acupuncture group,acupuncture and traditional acupuncture methods,once a day.The model group was not treated after operation,and the middle cerebral artery of the experimental rats was exposed and the thread was not embolized in the pseudo-operation group.The expression of GAP-43 protein in the surrounding tissues of cerebral ischemia was detected by neurological deficit score and immunohistochemistry at 3 days,7 days and 21 days after modeling.JD801 morphological image analysis system was used to analyze the results.Results:Each time period model The neurological function score of the group was significantly higher than that of the false operation group(P<0.05),and that of the other groups was significantly lower than that of the model group(P<0.05),and that of the acupuncture group,the traditional acupuncture group and the acupuncture combined with traditional acupuncture group was significantly lower than that of the model group(P<0.05),and the neurological deficit score of the other groups was significantly lower than that of the model group(P<0.05).On the 7th and 21st day after operation,compared with the traditional acupuncture group and the traditional acupuncture group,the neurological deficit score of the electro-acupuncture combined with traditional acupuncture group was significantly lower than that of the traditional acupuncture group(P<0.05).The immune expression of GAP-43 protein in brain tissue of model group was significantly higher than that of false operation group.The difference was statistically significant(P<0.05).Compared with the model group,the immune activity of GAP-43 in the electric acupuncture group,the traditional acupuncture group and the electro-acupuncture combined with traditional acupuncture group was significantly higher than that in the model group(P<0.05).On the 3rd,7th and 21st day after operation,compared with the traditional acupuncture group and the traditional acupuncture group,the immune activity of GAP-43 in the EA combined with traditional acupuncture group was significantly higher than that in the traditional acupuncture group(P<0.05).Conclusion:Acupuncture combined with traditional acupuncture can promote the brain tissue of rats with cerebral ischemia at all time points.The expression of GAP-43 improved the ischemic state of cerebral vessels.
