摘要:
目的:探讨电针改善小鼠脓毒症后认知功能障碍的作用及机制研究,为脓毒症的临床治疗及改善预后提供参考。方法:选择32只SD雄性小鼠作为研究对象,采用盲肠结扎穿孔法(GLP)对脓毒症小鼠模型进行制备,成功制备模型30只,之后将其随机分为脓毒症组(A组)、脓毒症+电针组(B组)、脓毒症+电针非经非穴组(C组)三组,各10只。其中B组和C组分别采用相同强度和频率的电针对“足三里”和“非经非穴”进行30 min刺激,而A组不进行处理。另取10只同批次健康小鼠作为对照组(D组)。于术后6 h对各组小鼠血清炎症因子水平进行比较,观察海马组织中丙二醛(MDA)、过氧化氢酶(CAT)及超氧化物歧化酶(SOD)水平差异,比较各组小鼠行为学指标及海马组织BDNF相对含量变化。结果:与D组相比,A组小鼠海马组织SOD、CAT水平及运动总距离均显著降低,海马组织MDA、血清TNF-α、IL-6、NSE水平以及中央格停留时间、场景性僵直时间均明显升高(均P<0.05); 与A组相比,B组小鼠上述指标明显改善(P<0.05); 且C组和A组两组小鼠上述指标比较无统计学差异(P>0.05)。结论:电针“足三里”可以有效的降低脓毒症小鼠术后炎症因子、应激反应指标的释放量,减轻脑部损伤,改善其认知功能,对脓毒症的临床治疗具有一定的指导意义。
Abstract:
Objective:To explore the effect and mechanism of electroacupuncture on improving cognitive dysfunction in mice after sepsis,and to provide reference for clinical treatment of sepsis and improving prognosis.Methods:Thirty-two SD male mice were selected as the research object,and the sepsis mouse model was prepared by cecal ligation and puncture method(GLP).30 models were successfully prepared.Afterwards,they were randomly divided into 3 groups of sepsis(group A),sepsis+electroacupuncture group(group B),sepsis+electroacupuncture non-acupuncture group(group C),10 each.Among them,group B and group C were stimulated by the same intensity and frequency for “Zusanli” and “Feijingfeixue” for 30 minutes,while group A was not treated.Another 10 healthy mice of the same batch were taken as the control group(Group D).The levels of serum inflammatory factors in mice of each group were compared at 6 hours postoperatively; the differences in the levels of malondialdehyde(MDA),catalase(CAT)and superoxide dismutase(SOD)in hippocampus were observed.The behavioral indexes of rats and the relative content of BDNF in hippocampus.Results:Compared with group D,SOD CAT level and total distance of exercise in hippocampal tissues of mice in group A were significantly decreased,MDA level of serum TNF-α,IL-6,NSE level in hippocampal tissues and time of central cell residence were significantly increased(all P<0.05).Compared with group A,the above indexes in group B were significantly improved(all P<0.05).There was no difference between group C and group A(P>0.05).Conclusion:Electroacupuncture Zusanli can effectively reduce the release of inflammatory factor stress response indexes after surgery in sepsis mice,reduce brain injury,and improve their cognitive function,which has certain guiding significance for the clinical treatment of sepsis.
