摘要:
目的:观察电针干预对炎性痛小鼠的镇痛作用及对前扣带皮层(ACC)HMGB1/TLR4信号通路相关蛋白的影响。方法:将健康C57BL/6小鼠随机分成五组,空白组(Control)、模型组(Model)、电针组(Model+EA)、抑制剂组(Model+GLY)、电针加抑制剂组(Model+GLY+EA),每组8只。除空白组外,其余各组均左侧踝关节注射完全弗氏佐剂(CFA)构建急性炎性痛模型。电针组取左侧“足三里”给予电针干预,每次20 min,1次/d,连续7 d。通过热敏痛、机械痛实验对各组小鼠机械痛阈值和热痛阈值测定,蛋白印迹法、免疫荧光法检测各组小鼠ACC脑区HMGB1/TLR4信号通路相关蛋白的变化。结果:与空白组相比,模型组小鼠热敏痛、机械痛实验中缩足反射潜伏期(PWL)明显缩短(P<0.001),小鼠ACC组织中HMGB1/TLR4信号通路相关蛋白HMGB1、TLR4、IL-1β表达水平显著增加(P<0.001),小鼠ACC脑区HMGB1蛋白表达显著增加。与模型组比较,电针组小鼠热刺痛、机械痛实验中PWL明显增加(P<0.001),小鼠ACC组织中HMGB1/TLR4信号通路相关蛋白HMGB1、TLR4、IL-1β表达水平显著下降(P<0.001),小鼠ACC脑区HMGB1蛋白表达显著下降。结论:电针干预可缓解CFA诱导的炎性痛,其机制可能与电针干预抑制HMGB1/TLR4信号通路及改善神经炎症有关。
Abstract:
Objective:To observe the analgesic effect of electroacupuncture on inflammatory pain mice and the effect of electroacupuncture on anterior cingulate cortex.The effect of HMGB1/TLR4 signaling pathway in Anterior cingulate cortex(ACC)was investigated.Methods:Healthy C57BL/6 mice were randomly divided into 5 groups:blank group(Control),Model group(Model),electroacupuncture group(Model+EA),inhibitor group(Model+GLY),electroacupuncture plus inhibitor group(Model+GLY+EA),8 mice in each group.Except control group,all the other groups were injected with complete CFA into the left ankle to construct acute inflammatory pain model.In the electroacupuncture group,the left “Zusanli” was taken for electroacupuncture intervention,20 min each time,for consecutive 7 days.The mechanical pain threshold and thermal pain threshold were measured by measurement of thermal thresholds and measurement of mechanical thresholds,and the changes of HMGB1/TLR4 signaling pathway related proteins in ACC brain region were detected by Western blot and Immunofluorescence method.Results:Compared with control group,PWL measurement of thermal thresholds and measurement of mechanical thresholds was significantly shortened in model group,and the expression levels of HMGB1,TLR4,IL-1β in HMGB1/TLR4 signaling pathway were significantly increased in ACC tissues of mice.The expression of HMGB1 protein in ACC brain region of mice was significantly increased.Compared with model group,PWL was significantly increased in measurement of thermal thresholds and mechanical pain experiments,and the expression levels of HMGB1,TLR4,IL-1β,HMGB1/TLR4 signaling pathway related proteins in ACC tissues of mice were significantly decreased in electroacupuncture group(P<0.001).The expression of HMGB1 protein in ACC brain region of mice was significantly decreased.Conclusion:Electroacupuncture intervention can alleviate CFA-induced inflammatory pain,and the mechanism may be related to the inhibition of HMGB1/TLR4 signaling pathway and the improvement of neuroinflammation.
