摘要:
目的:探讨扶金化积丸含药血清对肺癌A549细胞增殖及凋亡的影响及分子机制。方法:取SD大鼠40只,随机分为空白对照组和扶金化积丸组,空白对照组予等体积0.9%氯化钠溶液灌胃,给药组按生药量1.749 g/kg灌胃,提取并配制不同浓度含药血清(即150、75、37.5 mg/ml含药血清高、中、低剂量组)。A549细胞株复苏,取对数期细胞,以5×104/ml的密度接种至6孔培养板后继续培养至细胞贴壁,次日各实验组分别加入扶金化积丸高、中、低剂量含药血清500 μl/孔,待其干预细胞24 h后镜下观察细胞形态变化,拍照分析。MTT法检测含药血清对A549细胞的增殖抑制效应; 流式细胞术检测各组药物对细胞周期及凋亡的影响; Western blot法检测各组药物处理后凋亡蛋白Bax、Bcl-2的表达水平。结果:与空白对照组相比,扶金化积丸含药血清各剂量组均出现核固缩、圆细胞数量多且漂浮、胞质透亮且胞质内出现颗粒,细胞贴壁不牢,其中扶金化积丸含药血清高、中剂量组效果明显; 扶金化积丸高、中、低剂量组OD值较空白对照组明显减小(P<0.01或P<0.05); 扶金化积丸高、中剂量组G0/G1期A549细胞百分比显著增大,高剂量组S期、G2/M期A549细胞百分比明显减小(P<0.01或P<0.05); 扶金化积丸高、中、低剂量组A549细胞凋亡率均明显升高(P<0.01或P<0.05)。扶金化积丸高、中、低剂量组Bax表达及Bax/Bcl-2比例均上调(P<0.01),扶金化积丸高、中剂量组Bcl-2表达较空白对照组均下调(P<0.01或P<0.05)。结论:扶金化积丸含药血清能够抑制A549细胞的增殖,促进A549细胞凋亡,其机制可能与降低细胞中Bcl-2水平,增加Bax水平相关。
Abstract:
Objective:To investigate the effects of Fujin Huaji pill containing serum on the proliferation and apoptosis of lung cancer A549 cells and the possible molecular mechanism.Methods:A total of 40 SD rats were randomly divided into blank group and Fujin Huaji pill group.The blank group was given equal volume of normal saline by gavage,and the drug administration group was given gavage according to the drug dose of 1.749 g/kg.Serum containing different concentrations of SD was extracted and allocated to high,medium and low dose groups(150,75 and 37.5 mg/ml of SD serum).Cell line A549 was resuscitated,and the cells of log phase were inoculated into 6-well culture plates at a density of 5×104/ml,and then cultured until the cells stuck to the wall.The next day,each experimental group was added 500 μl/well of drug-containing serum in the high-dose,medium-dose and low-dose of Fujin Huaji pill,respectively.After 24 hours of intervention,the morphological changes of cells were observed under microscope and photographed for analysis.MTT assay was used to detect the proliferation inhibition effect of drug-containing serum on A549 cells.The effects of drugs on cell cycle and apoptosis were detected by flow cytometry.The expression levels of apoptotic proteins Bax and Bcl-2 were detected by Western blot.Results:Compared with the blank group,Fujin Huaji pill serum dose group showed nuclear pyretic,round cell number and floating,cytoplasm clear and granules in the cytoplasm,and cell adhesion was not firm,among which the effect of Fujin Huaji pill serum high and medium dose group was obvious.OD values in high,medium and low dose groups of Fujin Huaji pill were significantly decreased compared with blank control group(P<0.01 or P<0.05).The percentage of A549 cells in G0/G1stage was significantly increased in high and medium dose groups,and the percentage of A549 cells in S and G2/M stage was significantly decreased in high dose groups(P<0.01 or P<0.05).The apoptosis rate of A549 cells in high,medium and low dose groups of Fujin Huaji pill was significantly increased(P<0.01 or P<0.05).The expression of Bax and the proportion of Bax/Bcl-2 in serum high,medium and low dose groups of Fujin Huaji pill were up-regulated(P<0.01),and the expression of Bcl-2 in serum high and medium dose groups of Fujin Huaji pill was down-regulated compared with blank control group(P<0.01 or P<0.05).Conclusion:The serum containing Fujin Huaji pill can inhibit the proliferation of A549 cells and promote the apoptosis of A549 cells,which may be related to the decrease of Bcl-2 gene level and the increase of Bax gene level.
