摘要:
目的:观察针刺足三里对完全弗氏佐剂(CFA)诱导的炎性痛小鼠环磷酸鸟苷-腺苷酸合成酶(cGAS)/干扰素基因刺激因子(STING)信号通路表达的影响。方法:将C57BL/6J小鼠随机分为盐水组、模型组和针刺组,盐水组右后足底注射0.9%氯化钠溶液,模型组和针刺组右后足底注射CFA造模。针刺组选取足三里穴(ST36)针刺,盐水组与模型组同等固定,每日1次,共7次。测量小鼠热刺激缩足潜伏期(PWTL)、机械刺激缩足阈值(PWMT)以观察各组小鼠疼痛情况;采用RT-qPCR法检测背根神经节(DRG)和脊髓cGAS、STING、干扰素(IFN)-β的mRNA表达水平,结果:造模后,与盐水组相比,模型组小鼠PWTL、PWMT降低(P<0.05);针刺干预后,与模型组相比,针刺组PWTL、PWMT升高(P<0.05)。与盐水组相比,模型组小鼠DRG内cGAS、STING、IFN-β的mRNA表达水平降低(P<0.05),脊髓cGAS、STING、IFN-β的mRNA表达水平升高(P<0.05);与模型组相比,针刺组小鼠DRG内cGAS、STING、IFN-β的mRNA表达水平升高(P<0.05),脊髓cGAS、STING、IFN-β的mRNA表达水平下降(P<0.05)。结论:针刺能够改善CFA小鼠炎性痛;且在不同部位(DRG和脊髓)对cGAS/STING信号通路的mRNA表达具有双向调节作用。
Abstract:
Objective:To observe the effect of acupuncture on the expression of cyclic GMP-AMP synthase (cGAS) /stimulator of interferon genes (STING) signaling pathway in the spinal cord of inflammatory pain mice induced by Complete Freund’s Adjuvant (CFA).Methods:C57BL/6J mice were randomly divided into saline group,model group and acupuncture group.In the saline group,normal saline was injected into the right hind paw,and CFA was injected into the same hind paw of the model group and the acupuncture group.Acupuncture was applied to Zusanli (ST36) in the acupuncture group,and the other two groups were equally fixed,once a day for 7 times.Paw Withdrawal Thermal Latency (PWTL),Paw Withdrawal Mechanical Threshold (PWMT) and paw swelling were tested during the experiment.The mRNA expression of cGAS,STING,interferon(IFN)-β in spinal cord and dorsal root ganglion were detected by RT-qPCR.Results:Compared with the saline group,the PWTL and PWMT of model group was decreased (P<0.01).Compared with the model group,the PWTL and PWMT of acupuncture group was increased (P<0.01),until on the 7th day there was statistically difference between the two groups (P<0.01).Compared with the saline group,the mRNA expression of cGAS,STING,IFN-β in the dorsal root ganglion of the model group were decreased (P<0.05),and the mRNA expression of cGAS,STING,IFN-β in the spinal cord of the model group were increased (P<0.05).Compared with the model group,the expression of the above-mentioned genes of the acupuncture group were decreased (P<0.05).the expression of the above-mentioned genes of the acupuncture group were increased (P<0.05).Conclusion:Acupuncture can improve inflammatory pain in CFA mice;it also has a bidirectional regulatory effect on the mRNA expression of cGAS/STING signaling pathway in DRG and spinal cord.
