摘要:
目的:基于沉默信息调节因子1(SIRT1)/过氧化物酶体增殖受体γ共激活因子-1α(PGC-1α)通路探讨黄芪补肾活血汤对糖尿病肾病(DN)大鼠的保护作用及机制。方法:构建DN大鼠模型,成功建模的大鼠随机分成DN组、L-黄芪补肾活血汤组、M-黄芪补肾活血汤组、H-黄芪补肾活血汤组(灌胃11.46、22.92、45.84 g/kg黄芪补肾活血汤)、H-黄芪补肾活血汤+EX527组(灌胃45.84 g/kg黄芪补肾活血汤和腹腔注射5 mg/kg SIRT1/PGC-1α通路抑制剂EX527),每组各10只大鼠。另选10只正常大鼠为NC组。血糖仪检测大鼠血糖浓度;全自动生化分析仪检测大鼠血清肌酐(Scr)、血清胱抑素C(Cys-C)、尿素氮(BUN)水平;HE、Masson染色观察大鼠肾组织病理变化、纤维化情况;TUNEL染色检测大鼠肾组织细胞凋亡;ELISA检测大鼠血清MDA、SOD表达;DHE探针法检测大鼠肾组织ROS表达;Western blot检测SIRT1/PGC-1α信号通路相关蛋白表达。结果:DN组大鼠肾组织病理损伤和纤维化严重,血糖浓度、Scr、Cys-C、BUN、细胞凋亡率、MDA、ROS水平高于NC组(均P<0.05),SOD、SIRT1、PGC-1α表达低于NC组(均P<0.05);L-黄芪补肾活血汤、M-黄芪补肾活血汤、H-黄芪补肾活血汤组大鼠肾组织病理损伤和纤维化减轻,血糖浓度、Scr、Cys-C、BUN、细胞凋亡率、MDA、ROS水平低于DN组(均P<0.05),SOD、SIRT1、PGC-1α表达高于DN组(均P<0.05);H-黄芪补肾活血汤+EX527组大鼠肾组织病理损伤和纤维化加重,血糖浓度、Scr、Cys-C、BUN、细胞凋亡率、MDA、ROS水平高于H-黄芪补肾活血汤组(均P<0.05),SOD、SIRT1、PGC-1α表达低于H-黄芪补肾活血汤组(均P<0.05)。结论:黄芪补肾活血汤能够对DN大鼠肾组织产生保护作用,其机制可能与激活SIRT1/PGC-1α信号通路相关。
Abstract:
Objective:To explore the protective effect and mechanism of Huangqi Bushen Huoxue decoction on diabetic nephropathy (DN) rats based on the slient information regulator 1 (SIRT1)/peroxisome proliferator-activated receptor gamma coactivator-1α (PGC-1α) pathway.Methods:DN rat model was constructed.The successfully modeled rats were randomly separated into DN group,L-Huangqi Bushen Huoxue decoction group,M-Huangqi Bushen Huoxue decoction group,H-Huangqi Bushen Huoxue decoction group (orally administered 11.46、22.92、45.84 g/kg Huangqi Bushen Huoxue decoction),and H-Huangqi Bushen Huoxue decoction+EX527 group (orally administered 45.84 g/kg Huangqi Bushen Huoxue decoction and intraperitoneally injected 5 mg/kg SIRT1/PGC-1α pathway inhibitor EX527),there were 10 rats in each group.Another 10 normal rats were included as the NC group.The blood glucose meter was used to measure the blood glucose concentration in rats.Fully automated biochemical analyzer was used to test serum creatinine (Scr),serum cystatin (Cys-C),and blood urea nitrogen (BUN) in rats.HE and Masson staining were performed to observe the pathological changes and fibrosis of rat kidney tissue.TUNEL staining was used to test apoptosis in rat kidney tissue cells.ELISA was used to detect MDA and SOD in rat serum.The DHE probe method was performed to detect ROS in rat kidney tissue.Western blot was performed to measure the proteins related to the SIRT1/PGC-1α signaling pathway.Results:The pathological damage and fibrosis were severe in the renal tissue of DN group,with higher blood glucose concentration,Scr,Cys-C,BUN,apoptosis rate,MDA,and ROS (all P<0.05),and lower SOD,SIRT1,and PGC-1α than NC group (all P<0.05).The L-Huangqi Bushen Huoxue decoction group,M-Huangqi Bushen Huoxue decoction group,and H-Huangqi Bushen Huoxue decoction group showed reduced pathological damage and fibrosis in rat kidney tissue,with lower blood glucose concentration,Scr,Cys-C,BUN,apoptosis rate,MDA,and ROS (all P<0.05),and higher SOD,SIRT1,and PGC-1α than NC group (all P<0.05).The H-Huangqi Bushen Huoxue decoction+EX527 group showed aggravated renal tissue pathological damage and fibrosis,with higher blood glucose concentration,Scr,Cys-C,BUN,apoptosis rate,MDA,and ROS (all P<0.05),and lower SOD,SIRT1,and PGC-1α than NC group (all P<0.05).Conclusion:Huangqi Bushen Huoxue decoction have a protective effect on renal tissue of DN rats,which may be achieved by activating SIRT1/PGC-1α signaling pathway.
