摘要:
目的:探讨针刺对营养型肥胖小鼠肾周脂肪组织PHF23基因和Apelin/APJ信号通路的影响机制。方法:选择8周龄C57 BL/6小鼠80只,随机分为对照组、模型组、针刺组和APJ抑制剂组,各20只。对照组正常饮食,其他三组高脂饮食8周复制营养型肥胖模型。针刺组选择中脘、关元、天枢、足三里4个穴位。APJ抑制剂组在针刺前1周于尾静脉注射APJ抑制剂。4周后处死获取肾周脂肪组织,HE染色观察脂肪组织形态,qRT-PCR检测PHF 23基因,Western blot检测Apelin和APJ蛋白。检测T1(喂养前)、T2(喂养8周)、T3(针刺2周)和T4(针刺4周)时刻体重、总胆固醇(TC)、三酰甘油(TG)、低密度脂蛋白(LDL)、瘦素和脂联素水平。结果:与对照组比较,模型组肾周脂肪重量和脂肪细胞直径明显增加,而脂肪细胞数目明显减少(P<0.05);与模型组比较,针刺组肾周脂肪重量和脂肪细胞直径明显减少,而脂肪细胞数目明显增加(P<0.05)。与模型组比较,APJ抑制剂组上述改变不明显(P>0.05)。与对照组比较,模型组PHF23基因、Apelin和APJ蛋白表达量明显增加(P<0.05);与模型组比较,针刺组PHF23基因、Apelin和APJ蛋白表达量明显减少(P<0.05)。与模型组比较,APJ抑制剂组PHF23基因和Apelin蛋白表达量明显减少(P<0.05),但APJ蛋白表达量改变不明显(P>0.05)。与对照组比较,模型组T2、T3和T4时刻体重、TC、TG、LDL、瘦素和脂联素水平明显增加(P<0.05);与模型组比较,针刺组T3和T4时刻体重、TC、TG、LDL、瘦素和脂联素水平明显减少(P<0.05)。与模型组比较,APJ抑制剂组上述改变不明显(P>0.05)。结论:针刺可以明显降低营养型肥胖小鼠体重、血脂和脂肪因子水平,改善肾周脂肪组织的病理变化,可能与抑制PHF23基因以及Apelin/APJ 信号通路的表达有关,拮抗APJ蛋白表达可以抵消针刺的上述作用。
Abstract:
Objective:To explore the mechanism of acupuncture on PHF 23 gene and Apelin/APJ signaling pathway in perirenal adipose tissue of nutritional obese mice.Methods:Eighty 8-week-old healthy SPF C57 BL/6 mice were randomly divided into control group,model group,acupuncture group and APJ inhibitor group,with 20 mice in each group.The control group had normal diet,other three groups had high-fat diet for 8 weeks to duplicate nutritional obesity model.In acupuncture group,four acupoints including Zhongwan,Guanyuan,Tianshu and Zusanli were selected once a day,left and right wheels were exchanged for points.The APJ inhibitor group was injected with APJ inhibitor into tail vein one week before acupuncture.After 4 weeks,the mice were euthanized to obtain perirenal adipose tissue,morphology of adipose tissue was observed by HE staining,PHF 23 gene expression was detected by qRT-PCR,and Apelin and APJ protein expressions were detected by Western blot.Body weight,total cholesterol (TC),triglyceride (TG),low-density lipoprotein (LDL),leptin and adiponectin were measured at T1 (before feeding),T2 (8 weeks after feeding),T3 (2 weeks after acupuncture) and T4 (4 weeks after acupuncture)were detected.Results:Compared with control group,model group showed significant increases in perirenal fat weight and adipocyte diameter,while number of adipocyte decreased significantly (P<0.05).Compared with model group,acupuncture group showed significant decreases in perirenal fat weight and adipocyte diameter,while number of adipocyte increased significantly (P<0.05).Compared with model group,the above changes were not significant in APJ inhibitor group (P>0.05).Compared with control group,the levels of PHF 23 gene,Apelin,and APJ proteins in model group significantly increased (P<0.05).Compared with model group,PHF 23 gene,Apelin and APJ proteins in acupuncture group decreased significantly (P<0.05).Compared with model group,the expression levels of PHF 23 gene,Apelin protein significantly reduced in APJ inhibitor group (P<0.05),but APJ protein expression was not significant (P>0.05).Compared with control group,model group showed significant increases in body weight,levels of TC,TG,LDL,leptin and adiponectin at T2,T3,and T4 times (P<0.05).Compared with model group,acupuncture group showed significant decreases in body weight,levels of TC,TG,LDL,leptin and adiponectin at T3 and T4 times (P<0.05).Compared with model group,the above changes were not significant in APJ inhibitor group (P>0.05).Conclusion:Acupuncture can greatly reduce weight,blood lipid and adipokine levels of nutritional obese mice,improve pathological changes of perirenal adipose tissue,which may be related to inhibition of PHF23 gene and Apelin/APJ signal pathway expressions.Antagonistic APJ protein expression can counteract above effects of acupuncture.
