摘要:
目的:旨在探讨电针(EA)疗法是否通过环磷酸腺苷(cAMP)-蛋白激酶A(PKA)/激活转录因子2(ATF2)信号轴促进白色脂肪米色化产热,进而预防肥胖的发生。方法:将40只C57BL/6J小鼠随机分成四组,每组10只:正常(Ctrl)组、Model组、EA组和EA+H-89组。检测各组小鼠体重、Lee’s指数以及血脂指标含量。HE染色观察腹部脂肪组织的形态学变化。蛋白免疫印迹检测PR结构域包含蛋白16 (PRDM16)、解偶联蛋白1(UCP1)、过氧化物酶体增殖物激活受体γ共激活因子1α(PGC-1α)、PKA和ATF2蛋白表达水平。ELISA检测cAMP含量。结果:与Ctrl组比较,Model组的体重,Lee’s指数,总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)含量,白色脂肪质量显著增加;高密度脂蛋白胆固醇(HDL-C)含量、PRDM16、UCP1、PGC-1α、PKA和ATF2蛋白表达水平,cAMP含量显著降低(P<0.05)。与Model组比较,EA组的体重,Lee’s指数,TC、TG、LDL-C含量,白色脂肪质量显著降低;PRDM16、UCP1、PGC-1α、PKA和ATF2蛋白表达水平,cAMP含量显著增加(P<0.05)。与EA组比较,EA+H-89组的体重,Lee’s指数,TC、TG、LDL-C含量,白色脂肪质量显著增加;PRDM16、UCP1、PGC-1α、PKA和ATF2蛋白表达水平,cAMP含量显著降低(P<0.05)。结论:EA 能够激活cAMP-PKA/ATF2信号通路,进而上调白色脂肪组织米色化进程中的关键调控因子 PRDM16、UCP1以及 PGC-1α的表达水平,推动白色脂肪向米色脂肪的转化,增强机体的产热效能,发挥减肥降脂作用。
Abstract:
Objective:To investigate whether electroacupuncture (EA) therapy promotes beige thermogenesis of white fat through the cyclic adenosine monophosphate (cAMP)-protein kinase A (PKA)/activating transcription factor 2 (ATF2) signaling axis,thereby preventing the occurrence of obesity.Methods:40 C57BL/6J mice were randomly divided into 4 groups,10 in each group:Control (Ctrl) group,Model group,EA group and EA+H-89 group.The body weight,Lee’s index and blood lipid index of mice in each group were detected.HE staining was employed to observe the morphological alterations in abdominal adipose tissue.The protein expression levels of PR domain containing 16 (PRDM16),uncoupling protein 1 (UCP1),peroxisome proliferator-activated receptor gamma coactivator 1 alpha (PGC-1α),PKA and ATF2 were detected by Western blotting.The body weight,Lee’s index and blood lipcAMP was detected by ELISA.Results:Compared with the Ctrl group,the body weight,Lee’s index,total cholesterol (TC),triglyceride (TG),low-density lipoprotein cholesterol (LDL-C) content and white fat mass of the Model group were significantly increased.High-density lipoprotein cholesterol (HDL-C) content,PRDM16,UCP1,PGC-1α,PKA and ATF2 protein expression levels,and cAMP content were significantly reduced (P<0.05).Compared with the Model group,the body weight,Lee’s index,TC,TG,LDL-C content and white fat mass of the EA group were significantly decreased.The expression levels of PRDM16,UCP1,PGC-1 lipid index of mice in each cAMP content were significantly increased (P<0.05).Compared with the EA group,the body weight,Lee’s index,TC,TG,LDL-C content and white fat mass of the EA+H-89 group were significantly increased.The protein expression levels of PRDM16,UCP1,PGC-1roup were detected.cAMP content were significantly decreased (P<0.05).Conclusion:EA can activate the cAMP-PKA/ATF2 signaling pathway,and then up-regulate the expression levels of key regulatory factors PRDM16,UCP1 and PGC-1α in the process of white adipose tissue beige,promote the transformation of white fat to beige fat,enhance the body’s heat production efficiency,and play a role in weight loss and lipid reduction.
