摘要:
目的:探究针刺对血虚风燥型特应性皮炎(AD)豚鼠模型表皮通透屏障功能的调控机制。方法:将32只普通级纯白色雄性豚鼠随机分为模型组、空白组、对照组与针刺组,除空白组外,其余各组均采用2,4-二硝基氯苯丙酮溶液外涂诱导法建立血虚风燥型AD模型。模型组、空白组及针刺组均予以清水灌胃,针刺组在此基础上选取双肺俞、脾俞、膈俞、曲池等穴位,对照组予以氯雷他定片灌胃,均干预14 d。评价皮损情况[特应性皮炎面积及严重度指数评分(EASI)],检测屏障功能[皮肤角质层含水量(WCSC)、经皮水分流失量(TEWL)],ELISA法测定免疫指标(IgE、IL-4、IL-17)水平,Western blot法测定AQP3表达水平,逆转录聚合酶链反应(RT-PCR)检测皮损组织 AQP3 mRNA 表达情况。结果:与空白组比较,模型组EASI评分、TEWL与IgE、IL-4、IL-17水平均升高,差异具有统计学意义(P<0.05);WCSC与AQP3、皮损组织AQP3 mRNA表达水平均降低,差异具有统计学意义(P<0.05)。与模型组比较,针刺组EASI评分、TEWL与IgE、IL-4、IL-17水平均降低,差异具有统计学意义(P<0.05);WCSC与AQP3、皮损组织AQP3 mRNA表达水平均升高,差异具有统计学意义(P<0.05)。结论:针刺干预血虚风燥型AD豚鼠模型,确有改善表皮水分调节与表皮通透屏障功能的作用,其作用机制与调节免疫应激反应与AQP3、皮损组织AQP3 mRNA等表达水平相关。
Abstract:
Objective:To investigate the regulatory mechanism of acupuncture on epidermal permeability barrier function in a guinea pig model of atopic dermatitis (AD) with blood deficiency and wind-dryness pattern.Methods:32 ordinary-grade pure white male guinea pigs were randomly divided into a model group,blank group,control group,and acupuncture group.Except for the blank group,the other groups were used to establish an AD model with blood deficiency and wind-dryness pattern by topical application of 2,4-dinitrochlorobenzene (DNCB) acetone solution.The model group,blank group,and acupuncture group were given distilled water via gavage.Based on this,the acupuncture group received acupuncture at bilateral Feishu (BL 13),Pishu (BL 20),Geshu (BL 17),and Quchi (LI 11).The control group was administered loratadine tablets via gavage.All interventions lasted for 14 days.Skin lesion severity was evaluated [Eczema Area and Severity Index (EASI) score],barrier function was assessed [stratum corneum water content (WCSC),transepidermal water loss (TEWL)],immune markers (IgE,IL-4,IL-17) were measured by ELISA,AQP3 expression was detected by Western blot,and AQP3 mRNA expression in lesional skin tissue was measured by reverse transcription polymerase chain reaction (RT-PCR).Results:Compared with the blank group,the model group showed significantly increased EASI scores,TEWL,and levels of IgE,IL-4,and IL-17 (P<0.05),and significantly decreased WCSC,AQP3 expression,and AQP3 mRNA expression in lesional skin tissue (P<0.05).Compared with the model group,the acupuncture group showed significantly decreased EASI scores,TEWL,and levels of IgE,IL-4,and IL-17 (P<0.05),and significantly increased WCSC,AQP3 expression,and AQP3 mRNA expression in lesional skin tissue (P<0.05).Conclusion:Acupuncture intervention in the guinea pig model of AD with blood deficiency and wind-dryness pattern effectively ameliorated epidermal water regulation and epidermal permeability barrier function.Its mechanism of action is related to regulating the immune stress response and the expression levels of AQP3 and AQP3 mRNA in lesional skin tissue.
