摘 要 目的: 观察桃红四物汤对CCl4诱导小鼠肝纤维的治疗作用,并探究其可能机制。方法: 先将50只健康昆明小鼠随机取10只为正常对照组,余40只用30%CCl4花生油溶液按1 μl/g进行腹腔注射造模,造模完成后再随机分为模型组、秋水仙碱组(1 mg/kg)、桃红四物汤低、高剂量组(4.25 g/kg,17 g/kg),每组10只。造模后各组按剂量灌胃给药,1次/d,连续给药6周。末次给药后予以禁食不禁水,24h后心脏采血,分离血清,检测血清ALT、AST;取左叶肝脏,匀浆,取上清,检测HA、LN、PCⅢ、CⅣ、SOD、MDA、GSHPX含量;取右叶肝脏,4%多聚甲醛固定后HE染色进行病理学检查。结果:与模型组相比,桃红四物汤可明显降低CCl4诱导肝纤维模型中ALT、AST的含量,减少HA、LN、PCⅢ、CⅣ的释放,并能降低肝小叶结构破坏程度,减少肝细胞水肿、坏死、变性及炎症浸润,改善纤维组织增生及假小叶形成。同时,也可提高肝组织匀浆中SOD、GSHPX的活性,降低MDA的水平。结论:桃红四物汤对CCl4诱导肝纤维化模型具有保护作用,其保护作用机制可能是通过提高肝脏抗氧化的能力,降低脂质过氧反应实现的。
Abstract:
Abstract Objective:To investigate the effect of Taohong Siwu decoction CCl4induced on hepatic fibrosis model mice and potential mechanisms. Methods:50 healthy Kunming mice were randomly took 10 for normal control group,others were intraperitoneal injection of 30% on CCl4(1 μl/g) induce liver fibrosis model mice,then they were randomly divided into model group,colchicine group (1 mg/kg),Taohong Siwu decoction low  dose group(4.25 g/kg),high  dose group (17 g/kg). Then,every group was given the corresponding drug by gavage. After 6 weeks,to detect the activity of serum ALT,AST and the expression of HA,LN,PCⅢ,CⅣ,observe the results of HE staining and the expression of SOD,MDA,GSHPX. Results:Compared with the model group,Taohong Siwu decoction groups serum ALT,AST and HA,LN,PCⅢ,CⅣ were significantly decreased,HE staining results shown that the treatment groups of Taohong Siwu decoction can reduce the liver damage. At the same time,the expression of SOD,GSHPX were enhanced and MDA were decreased. Conclusion:Taohong Siwu decoction can be beneficial for CCl4 induced hepatic fibrosis model and it may be related to improve liver antioxidant capacity,reduce lipid peroxide reaction.
