摘要:
目的:观察电针干预对TBI大鼠脑组织结构形态、神经细胞凋亡及脑组织中p-GSK-3β、GLUT1蛋白表达的影响,以期探讨电针干预对TBI的治疗机理。方法:选择SD雄性大鼠76只,随机选取60只进行模型制备,造模成功后将其随机平均分为模型组和针刺组,预留假手术组和空白组各8只。于造模后3、7、14 d分批取材,HE染色观察损伤组织的形态结构; TUNEL法检测神经元细胞的凋亡情况; 免疫组化检测p-GSK-3β及GLUT1蛋白的表达情况。结果:HE染色可见造模后针刺组和模型组大鼠脑组织出现不同程度的坏死空洞区,损伤部位细胞排列紊乱,形态异常,但随着时间的推移,与模型组相比,针刺组坏死范围明显缩小,细胞形态及排列逐渐好转; TUNEL检测显示造模后模型组及针刺组出现大量的神经细胞凋亡,但随着时间的推移,针刺组及模型组的凋亡细胞数均有所减少,且针刺组减少更明显,差异具有统计学意义(P<0.05); 免疫组化检测p-GSK-3β及GLUT1蛋白的表达情况,结果显示在造模后3、7、14 d模型组及针刺组中两个目标蛋白的表达都呈增多趋势,且同时间点针刺组中p-GSK-3β及GLUT1蛋白的表达量均较模型组明显,差异具有统计学意义(P<0.05)。结论:电针可以减轻TBI大鼠脑组织的病理损伤程度及细胞凋亡数量,使脑组织中p-GSK-3β及GLUT1蛋白的表达量增加,发挥抗细胞凋亡的作用,减轻TBI后的神经继发性损害。
Abstract:
Objective:Observe the effects of electroacupuncture intervention on the brain tissue structure,nerve cell apoptosis and p-GSK-3β,GLUT1 protein expression in TBI rats,in order to explore the therapeutic mechanism of electroacupuncture intervention on TBI.Methods:Seventy-six male SD rats were selected,and 60 were randomly selected for model preparation.After successful modeling,they were randomly divided into a model group and an acupuncture group.A sham operation group and a blank group were reserved for 8 rats each.The materials were collected in batches at 3,7,and 14 d after modeling,and HE staining was used to observe the morphological structure of the injured tissue; TUNEL method was used to detect the apoptosis of neuronal cells; immunohistochemistry was used to detect the expression of p-GSK-3β and GLUT1 protein.Results:HE staining showed that the brain tissues of rats in the acupuncture group and the model group showed different degrees of necrotic cavities after the modeling,and the cells at the injury site were disorderly arranged and abnormal in shape,but with time,compared with the model group,necrosis The range was significantly reduced,and the cell morphology and arrangement gradually improved; TUNEL detection showed that a large number of neuronal apoptosis occurred in the model group and the acupuncture group after modeling,but the number of apoptotic cells in both the acupuncture group and the model group increased over time.Reduced,and the acupuncture group decreased more significantly(P<0.05); immunohistochemical detection of p-GSK-3β and GLUT1 protein expression,the results showed that after modeling 3,7,14 d model group and acupuncture group.The expression of the two target proteins in both showed an increasing trend,and the expression levels of p-GSK-3β and GLUT1 protein in the acupuncture group at the same time were significantly higher than those in the model group(P<0.05).Conclusion:Electroacupuncture can reduce the degree of pathological damage and the amount of apoptosis in the brain tissue of TBI rats,increase the expression of p-GSK-3β and GLUT1 protein in the brain tissue,play an anti-apoptotic effect,and reduce the neurological damage after TBI Hair damage.
