摘要:
目的:利用生物信息学技术探究山慈菇-王不留行在干预肺癌向骨转移过程中的相关作用机制。方法:运用中药系统药理学技术平台对山慈菇-王不留行的主要成分及作用靶基因进行筛选挖掘,在相关疾病数据库中筛选肺癌及骨转移瘤相关基因,映射三组靶基因得到关键靶点,通过String网站获得靶点PPI互相作用网络,根据Degree值筛选出核心靶点并通过DAVID数据库进行GO富集分析以及KEGG通路分析。结果:山慈菇-王不留行共包含6个有效成分,相对应靶点116个; 通过疾病数据库检索肺癌及骨转移瘤的已知疾病靶标,分别为1097、569个; 映射获得关键靶标35个,以Degree值≥20筛选核心靶点15个,富集出生物过程147条、信号通路66条。结论:山慈菇-王不留行一方面通过抑制体内血管生成构建、炎症因子释放,抑制肿瘤的生长发育; 另一方面通过干预破骨细胞等骨骼微环境,下调对转移肿瘤细胞的激活,阻碍肺癌骨转移过程。
