摘要:
目的:探索电针对颅脑损伤(TBI)大鼠脑组织中介导细胞存活的磷脂酰肌醇激酶/蛋白激酶B(PI3K/AKT)信号通路关键蛋白的影响。方法:选取雄性SD大鼠160只,随机分为空白组、假手术组、模型组、电针组、电针加阻断组、阻断剂组,采用改良Feeney自由落体撞击法制备TBI模型,取大鼠百会、水沟、内关、曲池、足三里、涌泉穴,于造模后24 h介入电针,共14 d。免疫荧光法检测TBI大鼠损伤脑组织PI3K、AKT蛋白的表达量。结果:电针后3、7、14 d,假手术组与空白组PI3K、AKT蛋白表达量比较,差异无统计学意义(均P>0.05)。模型组PI3K、AKT蛋白表达量低于假手术组、高于阻断剂组,差异有统计学意义(均P<0.05)。电针后3 d,电针组PI3K、AKT蛋白表达量与模型组比较,差异无统计学意义(均P>0.05)。电针组在7、14 d时PI3K、AKT表达量均高于模型组,差异有统计学意义(均P<0.05)。结论:电针可促进TBI大鼠损伤脑组织中PI3K、AKT蛋白表达,从而发挥对TBI大鼠脑损伤的康复效应。
Abstract:
Objective:To explore the effect of electroacupuncture on the key proteins of PI3K/AKT signaling pathway that mediate cell survival in brain tissue of rats with traumatic brain injury(TBI).Methods:160 male SD rats were selected and randomly divided into blank group,sham operation group,model group,electroacupuncture group,electroacupuncture combined blocking group and blocking agent group.The modified Feeney free-fall impact method was used to prepare the TBI model.Take the Baihui,Shuigou,Neiguan,Quchi,Zusanli,Yongquan points of the rat,and intervene with electroacupuncture 24 h after modeling for 14 days.The expression levels of PI3K and AKT protein in TBI rats were detected by immunofluorescence method.Results:Comparison of PI3K and AKT protein expression between 3,7,14 d after electroacupuncture,there was no difference between the sham operation group and the blank group(all P>0.05).The model group was lower than the sham operation group and higher than the blocker group, difference statistically significant(all P<0.05).There was no significant difference in PI3K and AKT protein expression between the electroacupuncture group and the model group 3 days after electroacupuncture(all P>0.05).The expression of PI3K and AKT in the electroacupuncture group was higher than those in the model group at 7,14 d,difference statistically significant(all P<0.05).Conclusion:Electroacupuncture can promote the expression of PI3K and AKT protein in the injured brain tissue of TBI rats,thereby exerting the rehabilitation effect on the brain injury of TBI rats.
