摘要:
目的:探讨八桂止痛散对带状疱疹后遗神经痛(PHN)大鼠脊髓背角组织JNK/p38 MAPK/NF-κB通路的影响。方法:取SD大鼠腹腔注射树脂毒素(RTX)制备PHN模型,随机分为对照组、模型组及八桂止痛散低、中、高剂量组,每组10只,另取10只SD大鼠作为对照组,以不同剂量八桂止痛散分组干预后检测大鼠长时程自发痛与痛觉过敏症状; 免疫荧光染色检测大鼠脊髓背角组织星形胶质细胞激活标记物GFAP与JNK/p38 MAPK/NF-κB p65通路相关蛋白阳性表达; ELISA检测大鼠脊髓背角组织与血清炎症因子水平; 免疫印迹检测大鼠脊髓背角组织JNK/p38 MAPK/NF-κB p65通路蛋白表达。结果:与对照组比较,模型组大鼠自发缩足反射次数、脊髓背角组织GFAP、p-JNK、p-p38 MAPK及p-NF-κB p65相对阳性表达、血清与脊髓背角组织白细胞介素-6(IL-6)及环氧化酶-2(COX-2)水平、脊髓背角组织p-JNK/JNK、p-p38 MAPK/p38 MAPK、p-NF-κB p65/NF-κB p65显著升高(P<0.05),机械痛阈值、热痛阈值降低(P<0.05); 低、中、高剂量八桂止痛散均可减轻PHN大鼠上述各病理指标变化(P<0.05),且呈剂量依赖性(P<0.05)。结论:八桂止痛散可抑制PHN大鼠脊髓背角组织中星形胶质细胞激活,进而抑制炎症反应,减轻大鼠自发痛与痛觉过敏症状,其机制可能与抑制JNK/p38 MAPK/NF-κB p65信号通路激活相关。
Abstract:
Objective:To investigate the effect of Bagui Zhitong powder on the JNK/MAPK/NF-κB pathway in the spinal dorsal horn tissue of rats with postherpetic neuralgia(PHN).Methods:SD rats were applied to prepare a PHN model by intraperitoneal injection of resin toxin(RTX).They were randomly separated into a control group,a model group,a low-dose Bagui Zhitong powder group,a medium dose Bagui Zhitong powder group,and a high-dose Bagui Zhitong powder group,with 10 rats in each group,another 10 SD rats as the control group,after intervention treatment with different doses of Bagui Zhitong powder,long-term spontaneous pain and hyperalgesia symptoms in rats were detected.The positive expression of GFAP activation markers and JNK/p38 MAPK/NF-κB p65 pathway related proteins in the spinal dorsal horn tissue of rats were detected by immunofluorescence staining.The levels of inflammatory factors in the spinal dorsal horn tissue and serum of rats were detected by ELISA.The expression of JNK/p38 MAPK/NF-κB p65 pathway related proteins in the spinal dorsal horn tissues of rats were detected by immunoblotting.Results:Compared with the control group,the number of spontaneous foot contraction reflexes,relative positive expression of GFAP,p-JNK,p-p38 MAPK,and p-NF-κB p65 in the spinal dorsal horn tissue,the levels of interleukin-6(IL-6)and cyclooxygenase-2(COX-2)in serum and spinal dorsal horn tissue,and p-JNK/JNK,p-p38 MAPK/p38 MAPK,p-NF-κB p65/NF-κB p65 in the spinal dorsal horn tissue of rats in the model group were obviously increased(P<0.05),the mechanical pain threshold and thermal pain threshold were obviously reduced(P<0.05).Low,medium,and high-doses of Bagui Zhitong powder can all alleviate the changes in the above pathological indicators in PHN rats(P<0.05),and were dose-dependent(P<0.05).Conclusion:Bagui Zhitong powder can inhibit the activation of astrocytes in the spinal dorsal horn tissue of PHN rats,thereby reducing the level of inflammatory response,alleviating spontaneous pain and hyperalgesia symptoms in rats which may be achieved by blocking the activation of the JNK/p38 MAPK/NF-κB p65 signaling pathway.
