摘要:
目的:基于同源性磷酸酶张力蛋白诱导激酶1(PINK1)/帕金森蛋白(Parkin)信号通路探讨丹黄散在糖尿病小鼠溃疡创面愈合中的作用。方法:选取18只SPF级雄性db/db小鼠,随机分为模型组、生长因子组、丹黄散组,每组6只,另选取6只SPF级雄性db/m小鼠作为空白组,各组小鼠均构建糖尿病溃疡模型。空白组与模型组小鼠以0.9%氯化钠溶液清创并用无菌纱布覆盖创面,生长因子组与丹黄散组小鼠在其基础上分别外敷表皮生长因子凝胶和丹黄散。观察比较各组小鼠溃疡创面愈合情况及PINK1/Parkin信号通路关键蛋白表达水平。结果:模型组第7、14、21天的创面愈合率明显低于丹黄散组、生长因子组(均P<0.05); HE染色和电镜观察创面组织,相较于模型组,生长因子组、丹黄散组创面肉芽组织病理改变明显减轻,大量成纤维细胞增生,致密分布,出现少量新生毛细血管,PINK1、Pakin和LC3-Ⅱ/Ⅰ蛋白表达水平均明显增加(均P<0.05)。结论:丹黄散可促进糖尿病小鼠溃疡创面愈合,可能通过调节PINK1/Pakrin信号通路进而调控线粒体自噬,保护线粒体功能,促进血管新生和肉芽组织生长,加快创面愈合。
Abstract:
Objective:To investigate the effect of Danhuang powder on ulcer wound healing in diabetic mice based on PINK1/Parkin signaling pathway.Methods:Eighteen SPF male db/db mice were randomly divided into model group,growth factor group and Danhuang powder group,6 mice in each group.Another 6 SPF male db/m mice were selected as blank group,diabetic ulcer models were constructed in each group.Blank group and model group were debrideed with 0.9% sodium chloride solution and covered with sterile gauze.Growth factor group and Danhuang powder group were applied to epidermal growth factor gel and Danhuang powder respectively.The ulcer wound healing and the levels of key proteins in PINK1/Parkin signaling pathway were observed and compared in each group.Results:The wound healing rate of model group at day 7,14 and 21 was lower than that of Danhuang powder group,growth factor group(P<0.05).Compared with model group,the pathological changes of granulation tissue in growth factor group and Danhuang powder group were significantly reduced.A large number of fibroblasts proliferated and distributed in a compact way and a few new capillaries appeared.Protein expression levels of PINK1,Pakin and LC3-Ⅱ/Ⅰwere significantly increased(all P<0.05).Conclusion:Danhuang powder can promote ulcer wound healing in diabetic mice,possibly regulate mitochondrial autophagy by regulating PINK1/Pakrin signaling pathway,protect mitochondrial function,promote angiogenesis and granulation tissue growth,and accelerate wound healing.
