摘要:
目的:探讨通心络胶囊干预转化生长因子-β1(TGF-β1)表达改善糖尿病心肌病(DCM)大鼠心肌纤维化损伤的作用机制。方法:将48只SPF级雄性大鼠随机分为对照组、模型组、缬沙坦组及通心络低、中、高剂量组,每组8只。除对照组外,其余大鼠均使用2%链脲佐菌素30 mg/kg一次性腹腔注射建立DCM模型。各组分别予以0.9%氯化钠溶液、缬沙坦及不同剂量通心络胶囊灌胃,连续干预4周。HE染色评估心脏损伤程度; Masson染色及免疫组化法评估心肌纤维化程度; ELISA法及Western blot法检测心肌组织TGF-β1蛋白的表达。结果:HE染色显示,与模型组相比,对照组大鼠心肌纤维形态均匀,排列规律紧密; 与对照组相比,模型组大鼠心肌纤维断裂,排列紊乱,不同剂量通心络组大鼠心肌纤维结构紊乱程度降低,心肌排列较模型组规则; Masson染色显示,模型组CVF高于对照组,各给药组CVF低于模型组; 免疫组化法、ELISA法及Western blot法检测显示,与对照组比较,模型组大鼠心肌组织TGF-β1蛋白表达升高(均P<0.05); 与模型组比较,各给药组TGF-β1蛋白表达降低(均P<0.05)。结论:通心络胶囊可能通过抑制TGF-β1蛋白表达改善DCM大鼠心肌纤维化和心肌结构紊乱。
Abstract:
Objective:To explore the mechanism of Tongxinluo capsule on improving the expression of transforming growth factor-β1(TGF-β1)and myocardial fibrosis in diabetic rats.Methods:48 SPF male rats were randomly divided into control group,model group,valsartan group and Tongxinluo low,middle and high dose groups,with 8 rats in each group.Except for the control group,the other rats were injected intraperitoneally with 2% streptozotocin 30 mg/kg to establish the model of diabetic cardiomyopathy.Each group was given 0.9% sodium chloride solution,valsartan and different doses of Tongxinluo capsule intragastric administration for 4 weeks.The degree of cardiac injury was evaluated by HE staining,the degree of myocardial fibrosis was evaluated by Masson staining and immunohistochemistry,and the expression of TGF-β1 protein in myocardial tissue was detected by ELISA and Western blot methods.Results:HE staining showed that compared with the model group,the shape of myocardial fibers in the control group was uniform and arranged closely.Compared with the control group,the myocardial fibers in the model group were broken and disordered,and the disorder of myocardial fiber structure in different doses of Tongxinluo group was lower,and the myocardial arrangement was more regular than that in the model group.Masson staining showed that the CVF in the model group was higher than that in the control group,and the CVF in each treatment group was lower than that in the model group.The results of immunohistochemistry,ELISA and Western blot showed that the expression of TGF-β1 protein in the model group was higher than that in the control group(all P<0.05),and the expression of TGF-β1 protein in each treatment group was lower than that in the model group(all P<0.05).Conclusion:Tongxinluo capsule may improve myocardial fibrosis and myocardial structural disorder in DCM rats by inhibiting the expression of TGF-β1 protein.
