目的:观察电针足三里、三阴交对糖尿病(DM)大鼠海马区胰高血糖素样肽1(GLP-1)的影响,探讨电针改善外周糖代谢、调控胰岛素的可能机制。方法:用随机数字表法将40只雄性SD大鼠随机分为空白组、模型组、对照组、电针组,每组10只。采用高脂饲料联合腹腔注射1%的STZ溶液按35 mg/kg进行造模。电针组对大鼠双侧足三里、三阴交电针,1次/d,20 min/次,连续干预21 d。对照组大鼠取胁下髂嵴上10 mm、后正中线旁开20 mm的固定对照点作为针刺部位,干预方法同电针组。定期测定大鼠空腹血糖水平,ELISA法检测血清胰岛素,Real-Time PCR法检测海马区GLP-1基因表达,Western blotting法检测海马区GLP-1蛋白表达。结果:与空白组比较,模型组和对照组大鼠空腹血糖上升,血清胰岛素水平、海马区GLP-1基因和蛋白表达下降,差异有统计学意义(均P<0.05)。与模型组比较,电针组大鼠空腹血糖下降,血清胰岛素水平、海马区GLP-1基因和蛋白表达上升,差异有统计学意义(均P<0.05)。结论:电针足三里、三阴交可以改善DM大鼠胰岛素分泌、降低外周血糖水平,并能提高海马区GLP-1基因和蛋白的表达,其机制可能与中枢调控胰岛素分泌有关。
Abstract:
Objective:To observe effect of electroacupuncture at Zusanli and Sanyinjiao on glucagon like peptide-1(GLP-1)in hippocampus of diabetes mellitus(DM)rats,and to explore the possible mechanism of synchronous improvement of improving peripheral glucose metabolism and regulateinsulin.Methods:40 male SD rats were randomly divided into blank group,model group,control group,and electroacupuncture group,10 rats in each group.The DM model was established by feeding hight-fat diet,and giving the intraperitoneal injection of 1% streptozotocin(STZ)by 35 mg/kg.The electroacupuncture group were treated with electroacupuncture intervention.A disposable sterile acupuncture needle was pierced into Zusanli and Sanyinjiao acupoints of bilateral hind limbs of rats.Electroacupuncture was performed 20 minutes per day,for 21 days in total.And in the control group,a fixed control point at 10 mm above the iliac crest and 20 mm next to the posterior midline was used as the acupuncture site,and the remaining operation was referred to the electroacupuncture group.The fasting blood glucose level of rats was measured regularly.The level of serum insulin was measured by ELISA.Real-Time fluorescence quantitative PCR and Western blotting were used to detect the expression of GLP-1 mRNA and protein in the hippocampus.Results:Compared with the blank group,the fasting blood glucose of rats were up-regulated,and the levels of serum insulin the GLP-1 mRNA and protein in the hippocampus were down-regulated in the model and control group,difference statistically significant(all P<0.05).Compared with the model group,the fasting blood glucose of the electro acupuncture group were lower and the levels of serum insulin the GLP-1 mRNA and protein in the hippocampus were higher,difference statistically significant(all P<0.05).Conclusion:Electroacupuncture at Zusanli and Sanyinjiao can improve insulin secretion in the DM rats and reduce peripheral blood glucose,raised GLP-1 mRNA and protein in the hippocampus.The mechanism may be related to the central regulation of insulin secretion.
