摘要:
摘 要 目的:探究紫苏子汤对哮喘小鼠肿瘤坏死因子α(TNFα)、白细胞介素8(IL8)、白介素1β(IL1β)表达的影响。方法:取60例Wistar小鼠,随机分为实验组和空白模型组,建立哮喘小鼠模型。实验组小鼠给予紫苏子汤,空白模型组小鼠给予生理盐水。使用Western blot法检测TNFα蛋白、IL8蛋白、IL1β蛋白表达,使用RTPCR法检测TNFα mRNA、IL8 mRNA、IL1β mRNA表达,使用全自动血液细胞分析仪检测中性粒细胞(Neutrophil)、淋巴细胞(Lymphocyte)、嗜酸性粒细胞(Eosinophil)计数,使用肺功能检测仪检测一秒用力呼气容积(FEV1)、2575%间强迫肺活量时用力呼气量(FEF2575)、呼气流量峰值(PEF)值。结果:实验组小鼠TNFα蛋白、IL8蛋白、IL1β蛋白表达水平均显著低于空白模型组小鼠(P<0.05);实验组小鼠TNFα mRNA、IL8 mRNA、IL1β mRNA表达水平均显著低于空白模型组小鼠(P<0.05);实验组小鼠中性粒细胞数显著高于空白模型组小鼠(P<0.05),实验组小鼠淋巴细胞数及嗜酸性粒细胞数明显低于空白模型组小鼠(P<0.05);实验组小鼠FEV1、FEF2575、PEF均明显高于空白模型组小鼠(P<0.05)。结论:紫苏子汤可以有效降低哮喘小鼠TNFα、IL8、IL1β水平,升高哮喘小鼠中性粒细胞数,降低哮喘小鼠淋巴细胞数及嗜酸性粒细胞数,修复哮喘小鼠受损肺功能。
Abstract:
Abstract Objective:To explore the effects of Zisuzi decoction on the expression of tumor necrosis factor  α (TNFα),interleukin 8 ( IL8) and interleukin 1 β ( IL1 β) in asthmatic mice.Methods:60 Wistar mice,were randomly divided into experimental group and blank model group to establish asthmatic mouse model. Mice in the experimental group were given Zisuzi decoction,and the blank model group was given normal saline. The expression of TNF α protein,IL8 protein and IL1 β protein was detected by western blot. The expression of TNF α mRNA,IL8 mRNA and IL1 β was detected by RTPCR,and the automatic blood cell analyzer was used to detect the count of neutrophils,lymphocytes,and eosinophilic granulocytes. Forced expiratory volume in one second (FEV1) and forced expiratory volume (Forced expiratory flow between 25) (Forced expiratory flow between) and peak expiratory flow value were detected by the lung function test instrument..Results:The expression level of TNF α protein,IL8 protein and IL1 β protein in the experimental group was significantly lower than that in the blank model group (P<0.05),and the expression level of TNF α mRNA,IL8 mRNA and IL1 β mRNA in the experimental group was significantly lower than that in the blank model group (P<0.05),and the number of neutrophils in the experimental group was significantly higher than that of the blank model mice (P<0.05) . The number of lymphocyte and the number of eosinophils in the experimental group were significantly lower than that in the blank model group (P<0.05),and the mice in the experimental group were significantly higher than that of the blank model group (P<0.05) in FEV1,FEF2575 and PEF.Conclusion: Zisuzi decoction can effectively reduce the level of TNF α,IL8 and IL1 β in asthmatic mice,increase the number of neutrophils in asthmatic mice,reduce the number of lymphocytes and eosinophils in asthmatic mice,and repair the damaged lung function of asthmatic mice.
