摘要:
目的:探讨透穴刺法对帕金森病焦虑模型大鼠杏仁核肿瘤坏死因子-α(TNF-α)、白细胞介素-10(IL-10)表达的影响。方法:60只大鼠采用盐酸6-羟基多巴(6-OHDA)损毁内侧前脑束制备帕金森病模型,造模成功后,大鼠随机分为正常组、模型组、黛力新组、透穴刺法组。正常组给予正常饲养,其余三组每日接受应激处理进一步诱导焦虑行为,黛力新组在每日应激处理前接受黛力新灌胃,透穴刺法组在每日应激处理前接受透穴刺干预。干预第15天时采用旷场实验和高架十字迷宫实验验证焦虑行为,随后采用Western blot实验检测大鼠杏仁核TNF-α、IL-10表达水平。结果:旷场实验中,模型组中央区停留时间百分比(RTDC)低于正常组,黛力新组、透穴刺法组RTDC高于模型组,且透穴刺法组RTDC高于黛力新组。高架十字迷宫实验中,模型组、黛力新组及透穴刺法组进入开放臂次数的百分比(PAOA)、开放臂停留时间百分比(RTDOA)均低于正常组。黛力新组、透穴刺法组的PAOA和RTDOA高于模型组,差异有统计学意义(均P<0.05),透穴刺法组PAOA和RTDOA高于黛力新组,差异有统计学意义(均P<0.05)。Western blot检测显示,模型组TNF-α表达水平高于正常组,IL-10表达水平低于正常组,黛力新组、透穴刺法组TNF-α表达水平低于模型组,透穴刺法组IL-10表达水平高于模型组、黛力新组。结论:帕金森病焦虑的发病机制可能与杏仁核TNF-α表达升高及IL-10表达下降有关,透穴刺法能够抑制TNF-α和促进IL-10表达,进而改善帕金森病焦虑症。
Abstract:
Objective:To investigate effect of acupoint piercing on the expression of tumor necrosis factor-α(TNF-α)and interleukin-10(IL-10)in amygdala of anxious rats with Parkinson's disease.Methods:60 rats were treated with 6-OHDA hydrochloride to damage the medial forebrain tract to establish Parkinson's model.The rats were randomly divided into normal group,model group,dalixin group and acupoint piercing group.The normal group was fed normally,and the other three groups received stimuli daily to further induce anxiety behavior.The delexin group received dalixin gavage before daily stress treatment,and the acupoint penetration acupuncture group received acupoint penetration intervention before daily stress treatment.The open field test and elevated cross maze test were used to verify the anxiety behavior at the 15th day after intervention,and then the expression levels of TNF-α and IL-10 in the amygdala of rats were detected by western blot.Results:In the open field experiment,the percentage of residence time(RTDC)in the central area of the model group was significantly lower than that of the normal group,the RTDC of the dalixin group and the acupoint penetration acupuncture group was significantly higher than that of the model group,and the RTDC of the acupoint penetration acupuncture group was higher than that of the dlixin group.In the elevated cross maze experiment,percentage of access the open arms(PAOA)and residence time distribution in the open arms(RTDOA)in the model group,the dalixin group and the acupoint piercing group were lower than those in the normal group.PAOA and RTDOA in the dalixin group and the acupoint piercing group were higher than those in the model group,differences statistically significant(all P<0.05).PAOA and RTDOA in the acupoint piercing group were higher than those in the dalixin group,differences statistically significant(all P<0.05).Western blot analysis showed that the expression of TNF-α in the model group was significantly higher than that in the normal group,and the expression of IL-10 was significantly lower than that in the normal group.The expression of TNF-α in the dalisin group and the acupoint penetration group was significantly lower than that in the model group,and the expression of IL-10 in the acupoint penetration group was significantly higher than that in the model group and the delexin group.Conclusion:The pathogenesis of Parkinson's disease anxiety may be related to the increased expression of TNF-α and the decreased expression of IL-10 in amygdala.Acupoint piercing can improve the anxiety of Parkinson's disease by inhibiting TNF-α and promoting the expression of IL-10 in amygdala.
