摘要:
目的:探讨桑芪滋阴补肾汤对高血压颈动脉粥样硬化发生和发展的影响及其作用机制。 方法:采用ox-LDL诱导HUVEC细胞建立血管内皮细胞损伤模型,分为空白对照组、模型组、高浓度桑芪滋阴补肾汤组、中浓度桑芪滋阴补肾汤组、低浓度桑芪滋阴补肾汤组和阳性药(阿托伐他汀)组。采用CCK8法检测各组细胞存活率; 采用RT-PCR法检测各组细胞中VEGFA的mRNA表达水平; 采用WB法检测各组细胞中NLRP3和Caspase-1的蛋白表达情况; 采用ELISA法检测肺泡灌洗液中IL-6和TNF-α水平。 结果:与空白对照组比较,模型组细胞存活率降低,细胞中VEGFA的mRNA表达水平降低,NLRP3和Caspase-1的蛋白表达水平升高,细胞上清液中IL-6、TNF-α的含量升高(P<0.05); 与模型组比较,低、中、高浓度桑芪滋阴补肾汤组细胞存活率升高,细胞中VEGFA的mRNA表达水平升高,NLRP3和Caspase-1的蛋白表达水平降低,细胞上清液中IL-6、TNF-α的含量降低,升高或降低水平与药物浓度呈正相关,差异具有统计学意义(P<0.05); 与阳性药组比较,高浓度桑芪滋阴补肾汤组细胞存活率、细胞中VEGFA的mRNA表达水平和NLRP3和Caspase-1的蛋白表达水平均无统计学差异(P>0.05)。 结论:桑芪滋阴补肾汤可以对高血压颈动脉粥样硬化发挥血管保护作用,其作用机制可能与通过抑制ox-LDL诱导细胞焦亡通路有关。
Abstract:
Objective:To explore the effect and mechanism of Sangqi Ziyin Bushen decoction on the occurrence and development of hypertensive carotid atherosclerosis.Methods:HUVEC cells were induced by ox-LDL to establish vascular endothelial cell injury model,which were divided into control group,model group,high concentration Sangqi Ziyin Bushen decoction group,medium concentration Sangqi Ziyin Bushen decoction group,low concentration Sangqi Ziyin Bushen decoction group and positive drug(atorvastatin)group.The cell survival rate of each group was detected by CCK8 method.The mRNA expression of VEGFA in each group of cells was detected by RT-PCR method.The protein expression of NLRP3 and Caspase-1 in cells of each group was detected by Western blotting method.Detection of IL-6 and TNF-α in bronchoalveolar lavage fluid by ELISA method.Results:Compared with the blank control group,the cell survival rate of the model group decreased,the mRNA expression of VEGFA decreased,the protein expression of NLRP3 and Caspase-1 increased,and the contents of IL-6 and TNF-α in the supernatant increased(P<0.05).Compared with the model group,the cell survival rate of Sangqi Ziyin Bushen decoction group increased in a dose-dependent manner,the mRNA expression of VEGFA in the cells increased,the protein expression levels of NLRP3 and Caspase-1 decreased,and the contents of IL-6 and TNF-α in the supernatant decreased,and they were positively correlated with drug concentration.The data were statistically significant(P<0.05).Compared with the positive drug group,there was no significant difference in cell survival rate,mRNA expression of VEGFA and protein expression of NLRP3 and Caspase-1 in high concentration Sangqi Ziyin Bushen decoction group(P>0.05).Conclusion:Sangqi Ziyin Bushen decoction can improve hypertensive carotid atherosclerosis and play a vascular protective effect,and its mechanism may be related to the inhibition of ox-LDL-induced cell death pathway.
